General conditions for lentiviral vector production
To produce lentivectors on demand for a collaborating group,
the following conditions have to be fulfilled prior to sending any material.
Viral vector production by Molecular Medicine (1 ml, 20-50 µl aliquots)
- Scientific collaboration: The collaborating laboratory is charged a fee (these are the net production costs 2006), which covers production of the LV (transfection reagents, vector production, vector purification and quality control (pg p24/ml, TU/ml (FACS)), consumables, labour). We provide packaging and envelope plasmids; transfer plasmid is provided at the proper purity and quality by the requesting group.
In addition, we require co-authorship on publication.
Viral vector cloning
We provide the transfer plasmid to the collaborating laboratory under MTA (after selecting the most appropriate transfer plasmid), and the latter carries out the cloning and sequencing procedures. We request quality control of the plasmid (sequencing of insert, restriction digest) prior to production of vector.
The transfer plasmid should be delivered to Molecular Medicine in endotoxin-free water and in sufficient amount (320 or 1600 μg for 2 or 10-layer cell-factory, respectively) (Gigaprep Sigma or Giga Qiagen).
Important:
- For all lentivector transfer plasmids, a material transfer agreement document has to be signed
- The shipping costs are covered by the requesting laboratory (dry ice shipment).
