Studying the role of LEDGF/p75 in integration site selection of HIV-1 and lentiviral vectors.

Function: 100% doctoral researcher at Division of Molecular Medicine from 1st Sep. 2008 for 1 year (renewable). The candidate has obtained a Master in Bioscience Engineering, Biomedical Sciences, Sciences.
We are looking for PhD candidates with a particular interest for both bioinformatics and virology.
Description of the project:
A hallmark of retroviral replication is the reverse transcription of the viral RNA genome into a DNA copy that becomes stably integrated into the host cell genome. The integration event forever links the fate of the invading retrovirus with that of the host cell. It is generally accepted that cellular proteins take part in the viral replication process and that integration into specific genomic sites depends on interaction with locally bound host transcription factors. The viral integrase (IN) plays a pivotal role. However, the underlying molecular mechanism for integration sites selection is still to be unveiled. In 2003 Lens Epithelial-Derived Growth Factor (LEDGF/p75) was characterized by our group as a novel and specific cellular binding partner of HIV IN, orchestrating nuclear accumulation and chromosomal localization of IN. The interaction of LEDGF/p75 and IN is lentiviral specific. Our group demonstrated the key role of LEDGF/p75 during HIV replication using RNAi. Taken together, current data provide strong clues for LEDGF/p75 to be the cellular co-factor responsible for the tethering of the HIV PIC to the chromatin and hence integration site selection of lentivirinae.
Objective: to understand the molecular principles of HIV integration target site selection using systems biology and bioinformatics approaches and to apply this knowledge for the design of safe viral vectors for gene therapy

1 Postdoctoral position in HIV-1 Molecular Virology (M/V)
A postdoctoral position is available immediately to join the Molecular Medicine Division at KULeuven University, Leuven, Flanders, Belgium, a unit dedicated to basic and applied research in HIV-1 biology and AIDS. HIV requires multiple cellular proteins to achieve its replication cycle. Our group has gained strong expertise during recent years in the identification, validation and characterization of co-factors of HIV integration (Van Maele et al. TiBS, 31(2): 98-105. (2006)). The project will focus on the further characterization of LEDGF/p75 as co-factor of HIV-1 integration and the discovery of new co-factors. The candidate will work in an interdisciplinary team of biochemists, virologists, cell biologists and biophysicists.
Requirements:
The requirements for the position are a doctoral degree; strong training and experience in cell biology and molecular biology; an excellent command of English; the desire to work within a team. The appointment will be for two years.

Omschrijving van de functie :
Binnen onze dynamische academische onderzoeksgroep zoeken wij een gemotiveerde laborant(e) om, in samenwerking met doctorale en postdoctorale onderzoekers, wetenschappelijke onderzoeksexperimenten uit te voeren in het kader van de ziekte van Parkinson (pathogenese, gentherapie, stamceltherapie en moleculaire beeldvorming) met behulp van lentivirale vectoren.
Specifieke opdrachten :
- Neurochirurgische stereotactische operaties op kleine proefdieren (muis en rat).
- Uitvoeren van gedragstesten op proefdieren
- Histologie, immunohistochemie en microscopie
- Moleculair-biologisch werk en eiwitanalyses, onder andere: RNA- en DNA-technologie, cloneringstechnieken, PCR-technieken, Western blot.
- Celcultuur
- Verantwoordelijkheid voor de organisatie en toezicht op proefdierexperimenten, voor het instrumentarium, en voor het naleven van de specifieke reglementering
Profiel
- Kandidaten zijn minstens in het bezit van een diploma hoger onderwijs van het korte type, bij voorkeur richting farmaceutische en biologische technieken.
- Kandidaten hebben een grondige interesse en inzicht in het verloop van wetenschappelijke experimenten.

De Afdeling Moleculaire Geneeskunde is op zoek naar aspirant-doctoraatsstudenten die in aanmerking komen voor een aanvraag om een specialisatiebeurs bekomen van het IWT met de bedoeling het behalen van een doctoraat in de medische wetenschappen. Het onderwerp van de aanvraag zal betrekking hebben op cellulaire co-factoren van de HIV-replicatie. De doctoraten zouden moeten leiden naar een beter begrip van hun rol in de virale replicatie cyclus en hun betekenis voor de ontwikkeling nieuwe antivirale doelwitten.
Profiel: laatste jaar Biomedische Wetenschappen, Biologie, Bio-ingenieur, Biotechnologie, Arts, Dierenarts met zeer goede studieresultaten in de afgelopen jaren.

Task description:

• Strategic planning of R&D in collaboration with different principle investigators.
Requirements:
• Experience with molecular biology is a necessity. Interest in molecular imaging.

Doel van de functie
