Tryptophan-mediated photolysis of disulfide bonds in proteins and peptides.

Recently we presented evidence of Trp-mediated photolysis of disulfide bonds within goat alpha-lactabumin (GLA) upon irradia- tion of the protein at 280 or 295 nm. In this project (a) we will investigate the consequences of the Trp-mediated photolysis for the physical-chemical properties and for the biological function of GLA. We also will investigate whether GLA can gain resistance against illumination by substitution of one or more Trp- for non-chromophoric groups. (b) The impact of the factors that govern Trp-mediated photolysis by near-UV light will be studied in photolysis experiments with synthetic, cyclic peptides containing a single Trp residue and one disulfide bridge which closes the circle. (c) We intend to extend our study to other proteins for which the photolysis of a disulfide bond may affect their biological activity.
