gestational diabetes melitus: studies in humans and animal models on the role of obesity and adipokines.

Obesity is the most important risk factor for the development of gestational diabetes(GDM). The offspring of mothers with GDM have an increased risk for developing obesity and glucose intolerance, which is not entirely explained by genetic factors, but to some degree initiated in utero. In this project,we study the interplay between genetic and environmental factors in association with obesity and GDM, and glucose tolerance in the offspring. In preg- nant women, we measure adipokines as well as genetic variation of leptin receptor and PPARg in relation to body weight/weight gain during pregnancy and the development of GDM. In rats, we found that exogenous obesity through a hypercaloric diet induces GDM.We will study the relation between adipokine gene expression in adi- pose tissue, placenta and pancreatic islets, insulin sensitivity and beta-cell function. In the adult offspring,we measure glucose tolerance, insulin sensitivity and adipokine gene expression.
